
Maolan inhibits the calcium channel regulatory activity of TMBIM6 by specifically binding to it, leading to endoplasmic reticulum calcium homeostasis imbalance and ROS accumulation in ESCC cells. This process induces strong endoplasmic reticulum stress (ER Stress) and immunogenic cell death (ICD), prompting tumor cells to release "danger signals" (CRT, HMGB1, ATP), thereby promoting dendritic cell maturation and CD8+ T cell infiltration and killing, ultimately reshaping the immunosuppressive microenvironment of ESCC.
